Gwynne began her career as an actress in her twenties. In 1989, she starred as feminist lecturer Dr. Robyn Penrose in the BBC television mini-series version of David Lodge’s Nice Work.

In 1990, she had her first high-profile comedy role as Alex Pates in Drop the Dead Donkey. She then appeared in the 1991 Children’s ITV science-fiction series Time Riders and later became a regular in Peak Practice, first debuting as Dr Joanna Graham at the start of Series 7 in 1999. In 2002, she played Emma Faraday in the BBC television drama The Secret.

From the Octagon in Bolton in Hedda Gabler to Richard Cheshire’s Way of the World, she appeared in West End productions of Ziegfeld as Billie Burke (1988), City of Angels, and Billy Elliot the Musical at the Victoria Palace Theatre, for which she was nominated for an Olivier Award.

Gwynne also appeared in a number of Royal Shakespeare Company plays. Her later television performances were typically in shorter plays, such as her role as Julius Caesar’s wife, Calpurnia, in the television series Rome.
